چکیده

IVF success rates are low, with approximately one-third of all cycles resulting in live births1. Improving IVF success rates would make a significant difference in the lives of many women trying to conceive. Chromosome aneuploidy (abnormal number of chromosomes) is a major cause of IVF failure, miscarriage, and, in rare cases, abnormal pregnancy and live birth. Most aneuploidies arise during the final stages of egg development and increase exponentially with maternal age. Selectively implanting euploid (normal number of chromosomes) embryos is critical to reducing miscarriage and improving IVF success. Early clinical experience show improvements as high as 50% in IVF success rates using embryos selected by the 24sure test2.

I-37: Establishing High Resolution Genomic Profiles of Single Cells Using Microarray and Next-Generation Sequencing Technologies

The nature and pace of genome mutation is largely unknown. Standard methods to investigate DNA-mutation rely on arraying or sequencing DNA from a population of cells, hence the genetic composition of individual cells is lost and de novo mutation in cell(s) is concealed within the bulk signal. We developed methods based on (SNP-) arraying and next-generation sequencing of single-cell whole-genom...

Towards a Next-Generation Sequencing Diagnostic Service for Tumour Genotyping: A Comparison of Panels and Platforms

Detection of clinically actionable mutations in diagnostic tumour specimens aids in the selection of targeted therapeutics. With an ever increasing number of clinically significant mutations identified, tumour genetic diagnostics is moving from single to multigene analysis. As it is still not feasible for routine diagnostic laboratories to perform sequencing of the entire cancer genome, our app...

Bioinformatic Challenges in Clinical Diagnostic Application of Targeted Next Generation Sequencing: Experience from Pheochromocytoma

BACKGROUND Recent studies have demonstrated equal quality of targeted next generation sequencing (NGS) compared to Sanger Sequencing. Whereas these novel sequencing processes have a validated robust performance, choice of enrichment method and different available bioinformatic software as reliable analysis tool needs to be further investigated in a diagnostic setting. METHODS DNA from 21 pati...

PIPETMAX®: Automation of the Illumina® Nextera® XT DNA Library Preparation Kit

The Illumina® Nextera® XT DNA Library Preparation Kit workflow was automated on PIPETMAX®. For comparison, 12 technical replicate libraries were prepared side by side, using manual liquid handling or automated on PIPETMAX. The 24 libraries were pooled and sequenced on an Illumina MiSeq® sequencing system. Both library preparation methods generated high quality data with >95% mapped reads and op...

Towards Clinical Molecular Diagnosis of Inherited Cardiac Conditions: A Comparison of Bench-Top Genome DNA Sequencers

BACKGROUND Molecular genetic testing is recommended for diagnosis of inherited cardiac disease, to guide prognosis and treatment, but access is often limited by cost and availability. Recently introduced high-throughput bench-top DNA sequencing platforms have the potential to overcome these limitations. METHODOLOGY/PRINCIPAL FINDINGS We evaluated two next-generation sequencing (NGS) platforms...
